ABSTRACT

It is unclear whether physical activity and cardiorespiratory fitness (CRF) are pathways that link low pulmonary function (LPF) to increased blood pressure (BP). Therefore, we investigated the extent to which CRF and moderate-to-vigorous physical activity (MVPA) mediate the relationship between LPF and high BP in adults. We conducted a cross-sectional study with 1,362 participants that underwent cardiopulmonary exercise testing (CPET), spirometry, and wore an accelerometer to determine physical activity patterns. We performed mediation analyses using structural equations considering peak oxygen uptake (V̇O2) and MVPA as mediators, forced vital capacity (FVC) and forced expiratory volume in the first second (FEV1) as independent variables, and systolic and diastolic blood pressure (SBP, DBP) as dependent variables. The probability of alpha error was set at 5%. We found a significant total effect of FVC on SBP and DBP considering V̇O2 as mediator (P<0.01). Indirect effects were also significant, with 42.6% of the total effect of FVC on SBP and 77% on DBP mediated by V̇O2 (P<0.01). We did not observe a direct effect of FVC on SBP and DBP. Considering FEV1 as an independent variable, the total effect on SBP was also significant, as were the indirect effects, mediated by V̇O2 at 14.8% for SBP and 7.6% for DBP (P<0.01). We did not find an indirect effect of FVC or FEV1 considering the MVPA as a mediator. CRF mediates the pathway that links LPF and elevated BP. Therefore, CRF is more sensitive to variations in FVC and FEV1 than MVPA.

ABSTRACT

Epidermal growth factor receptor (EGFR) signaling and components of the fibrinolytic system, including urokinase-type plasminogen activator (uPA) and thrombomodulin (TM), have been implicated in tumor progression. In the present study, we employed cBioPortal platform (http://www.cbioportal.org/), cancer cell lines, and an in vivo model of immunocompromised mice to evaluate a possible cooperation between EGFR signaling, uPA, and TM expression/function in the context of cervical cancer. cBioPortal analysis revealed that EGFR, uPA, and TM are positively correlated in tumor samples of cervical cancer patients, showing a negative prognostic impact. Aggressive human cervical cancer cells (CASKI) presented higher gene expression levels of EGFR, uPA, and TM compared to its less aggressive counterpart (C-33A cells). EGFR induces uPA expression in CASKI cells through both PI3K-Akt and MEK1/2-ERK1/2 downstream effectors, whereas TM expression induced by EGFR was dependent on PI3K/Akt signaling alone. uPA induced cell-morphology modifications and cell migration in an EGFR-dependent and -independent manner, respectively. Finally, treatment with cetuximab reduced in vivo CASKI xenografted-tumor growth in nude mice, and decreased intratumoral uPA expression, while TM expression was unaltered. In conclusion, we showed that EGFR signaling regulated expression of the fibrinolytic system component uPA in both in vitro and in vivo settings, while uPA also participated in cell-morphology modifications and migration in a human cervical cancer model.

ABSTRACT

El objetivo fue comparar la rugosidad de un composite nanoparticulado con diferentes técnicas de acabado y pulido. Se obtuvieron 20 discos en conformadores ad-hoc en los que se insertó un composite nanoparticulado en capas de 2mm. Se activó cada incremento durante 30s con una intensidad de 1450 mw/cm2. Todas las probetas se pulieron con la secuencia completa de discos Sof-lex a velocidad media y luego se asignaron a uno de 4 grupos (n=5) tratados de la siguiente manera: G1: sin tratamiento extra; G2: con cepillos Astrobrush y JC-SICRA-G; G3: con pasta de diamantes Diamond Gloss aplicada con fieltro; y pasta de óxido de aluminio Poligloss y fieltro; G4: igual que G3 + cepillos (igual que G2). Las superficies se evaluaron con un perfilómetro óptico tridimensional. Se registró la rugosidad promedio (Ra) en tres puntos de cada espécimen, se consideró la Ra a la media de los tres registros obtenidos. Los datos se analizaron mediante ANOVA de una vía y prueba de Tukey para comparaciones múltiples. Los resultados obtenidos fueron: G1: 132,20nm (26,16); G2: 77,20 nm (20,64); G3: 78,74 nm (10,04); G4: 66,47nm (31,08). Se encontró diferencia estadísticamente significativa entre los grupos (P=0.0017); de las comparaciones múltiples surge que G1 presentó una rugosidad media significativamente mayor que los restantes. G1>G2 (P<0.01), G1>G3 (P<0,05), G1>G4 (P<0,01). En conclusión, el uso de sistemas anexos de pulido permite lograr mayor lisura superficial. El uso de más de un sistema complementario no mejora el resultado (AU)

ABSTRACT

Neurological diseases are responsible for approximately 6.8 million deaths every year. They affect up to 1 billion people worldwide and cause significant disability and reduced quality of life. In most neurological disorders, the diagnosis can be challenging; it frequently requires long-term investigation. Thus, the discovery of better diagnostic methods to help in the accurate and fast diagnosis of neurological disorders is crucial. Circulating nucleic acids (CNAs) are defined as any type of DNA or RNA that is present in body biofluids. They can be found within extracellular vesicles or as cell-free DNA and RNA. Currently, CNAs are being explored as potential biomarkers for diseases because they can be obtained using non-invasive methods and may reflect unique characteristics of the biological processes involved in several diseases. CNAs can be especially useful as biomarkers for conditions that involve organs or structures that are difficult to assess, such as the central nervous system. This review presents a critical assessment of the most current literature about the use of plasma and serum CNAs as biomarkers for several aspects of neurological disorders: defining a diagnosis, establishing a prognosis, and monitoring the disease progression and response to therapy. We explored the biological origin, types, and general mechanisms involved in the generation of CNAs in physiological and pathological processes, with specific attention to neurological disorders. In addition, we present some of the future applications of CNAs as non-invasive biomarkers for these diseases.

ABSTRACT

The aim of this study was to assess the effects of chemical restraint, general anesthesia and opioid treatment on hematological components in Cuniculus paca. Eight healthy, adult, captivity female animals , underwent three laparoscopic procedures with a 15-day interval were evaluated. After physical restraint, an association of ketamine (25mg/kg) and midazolam (0.5mg/kg) was administered intramuscularly for chemical restraint. Posteriorly, anesthesia was induced and maintained with isoflurane; and randomly administered methadone (0.5mg/kg), tramadol (5mg/kg) or saline-placebo (0,1mL/kg) intramuscularly. After pharmacological restraint and in the final laparoscopy stage, venous blood samples were obtained for complete blood count, total plasma protein (TP), creatinine, alanine aminotransferase (ALT), sodium, potassium, chloride and ionized calcium analysis. During general anesthesia, hemoglobin, TP concentration and lymphocytes decreased (P=0.029; <0.001; 0.022 respectively), whereas the potassium levels increased (P=0.034). In conclusion, chemical restraint with ketamine/midazolam association causes a slight decrease in blood cellular components. Isoflurane anesthesia for laparoscopic procedure lead to decrease in hemoglobin, lymphocytes and protein concentrations, while potassium increased, without any influence from the tramadol or methadone treatment. However, these alterations were transient, and its hematologic values can collaborate in carrying out epidemiological, pathophysiological or case studies in the Cuniculus paca.(AU)


O objetivo do presente estudo foi avaliar os efeitos de contenção química, anestesia geral e tratamento com opiáceos nos parâmetros hematológicos em Cuniculus paca. Foram avaliados oito animais saudáveis, fêmeas, adultas, de cativeiro, que foram submetidas a três procedimentos laparoscópicos, com intervalo de 15 dias. Após a contenção física, uma associação de cetamina (25mg/kg) e midazolam (0,5mg/kg) foi administrada por via intramuscular para contenção química. Posteriormente, a anestesia foi induzida e mantida com isoflurano, e administrou-se aleatoriamente metadona (0,5mg/kg), tramadol (5mg/kg) ou placebo salina por via intramuscular. Após a contenção farmacológica e em estágio final da laparoscopia, foram obtidas amostras de sangue venoso para contagem sanguínea completa, proteína de plasma total (TP), creatinina, alanina aminotransferase (ALT), cálcio, sódio, potássio e cloreto ionizado. Durante a anestesia geral, a concentração de hemoglobina, TP e linfócitos diminuiu (P= 0,029;< 0,001; 0,022, respectivamente), enquanto os níveis de potássio aumentaram (P= 0,034). Em conclusão, a contenção química com associação de cetamina/midazolam promove uma ligeira diminuição dos componentes celulares do sangue. A anestesia com isoflavano para o procedimento laparoscópico levou a uma diminuição das concentrações de hemoglobina, linfócitos e proteínas, enquanto o potássio aumentou, sem qualquer influência do tratamento com tramadol ou metadona. No entanto, essas alterações foram transitórias, e os seus valores hematológicos obtidos podem colaborar na realização de estudos epidemiológicos, fisiopatológicos ou casuísticas para Cuniculus paca.(AU)

ABSTRACT

Aiming to provide cardiovascular morphophysiology information on the Cuniculus paca, an important neotropical rodent, eight healthy adult females of this species were evaluated three times by echocardiography under general anesthesia with isoflurane every 15 days. The exams were performed by a single experienced evaluator with the animals positioned in right and left decubitus. Posteriorly, two expert evaluators measured the cardiac chambers, walls and flow patterns, by B-mode, M-mode, and Doppler ultrasonography. The resulting values were compared among evaluators and periods by the Bland-Altman agreement test and several descriptive statistics were presented for each parameter. Echocardiographic images were obtained between the second and fifth left and right intercostal spaces, enabling the measurement of heart chambers and walls, mitral, tricuspid, aortic and pulmonary valves blood flows, and the ejection and shortening fractions calculation. None of the studied variables showed inter-observers or inter-periods variations. This study provided some normal echocardiographic variables, applicable to epidemiological, pathophysiological or case studies in the Cuniculus paca and phylogenetically close species.(AU)


Com o objetivo de fornecer informações da morfofisiologia cardiovascular da Cuniculus paca, um importante roedor neotropical, oito fêmeas adultas saudáveis dessa espécie foram avaliadas três vezes pela ecocardiografia, sob anestesia geral com isoflurano, a cada 15 dias. Os exames foram realizados por um único avaliador experiente, com os animais posicionados em decúbito direito e esquerdo. Posteriormente, dois avaliadores experientes mediram as câmaras cardíacas, as paredes e os padrões de fluxo, por meio do modo B, do modo M e da ultrassonografia Doppler. Os valores resultantes foram comparados entre avaliadores e períodos pelo teste de concordância de Bland-Altman, e várias estatísticas descritivas foram apresentadas para cada parâmetro. As imagens ecocardiográficas foram obtidas entre o segundo e o quinto espaços intercostais esquerdo e direito, o que permitiu a medição das câmaras e paredes cardíacas, do fluxo sanguíneo pelas válvulas mitral, tricúspide, aórtica e pulmonar e o cálculo das frações de ejeção e encurtamento. Nenhuma das variáveis estudadas mostrou variações entre os avaliadores ou entre os períodos. Este estudo fornece algumas variáveis ecocardiográficas normais, aplicáveis a estudos epidemiológicos, fisiopatológicos ou de casos na Cuniculus paca e em espécies filogeneticamente próximas.(AU)

ABSTRACT

Cervical cancer is a public health problem and the molecular mechanisms underlying radioresistance are still poorly understood. Here, we evaluated the modulation of key molecules involved in cell proliferation, cell cycle and DNA repair in cervical cancer cell lines (CASKI and C33A) and in malignant tissues biopsied from 10 patients before and after radiotherapy. The expression patterns of epidermal growth factor receptor (EGFR), excision repair cross-complementation group 1 (ERCC1) and p53 were evaluated in cancer cell lines by quantitative PCR and western blotting, and in human malignant tissues by immunohistochemistry. The mutation status of TP53 gene was evaluated by direct sequencing. Among cell lines, absent or weak modulations of EGFR, ERCC1 and p53 were observed after exposure to 1.8 Gy. Conversely, increased expressions of p53 (5/10 patients; P=0.0239), ERCC1 (5/10 patients; P=0.0294) and EGFR (4/10 patients; P=0.1773) were observed in malignant tissues after radiotherapy with the same radiation dose. TP53 mutations were found only in one patient. Here we show that a single dose of radiotherapy induced EGFR, ERCC1 and p53 expression in malignant tissues from cervical cancer patients but not in cancer cell lines, highlighting the gap between in vitro and in vivo experimental models. Studies on larger patient cohorts are needed to allow an interpretation that an upregulation of p53, EGFR and ERCC1 may be part of a radioresistance mechanism.

ABSTRACT

The aim of this study was to evaluate of the efficacy of PRP employment associated with surgical sponges to improve the integration of the graft in the recipient bed. It was held at the Veterinary Hospital UNESP, Campus of Jaboticabal - SP, a study of 64 rabbits, divided into eight groups with eight animals. The groups were divided in control with saline solution 0,9%, control with PRP both without the sponge, surgical sponge with PRP, surgical sponge without PRP, and were used mesh and layer grafts in the respective groups. The data were submitted to statistical analysis (paired t-test, Kruskal-Wallis test, with subsequent use of the multiple comparison tests of Dunn, analysis of variance (F) test, Tukey test, P< 0.05). Edema and exudate with 3 and 3 and 7 days (P= 0,03 e P= 0,0049); coloring on the 14th day (P= 0,0001); cosmetic appearance on the 7th and 14th day (P= 0,0026 and P= 0,0001); mononuclear cells (P= 0,01) and polymorphonuclear (P= 0,01); fibroblast proliferation (P= 0,01); collagenous (P= 0,05); hemorrhage (P-007); necrosis and re-epithelialization (P= 0,2928 and P= 0,1). We concluded that the use of Platelet Rich Plasma Gel on skin grafts associated with a sponge as a compressive dressing promote the skin graft survival without a previous granulation tissue.(AU)


O objetivo deste estudo foi avaliar a eficácia do PRP associado com esponjas cirúrgicas na integração do enxerto ao leito receptor. Realizou-se, no Hospital Veterinário da Unesp, Jaboticabal, SP, um estudo com 64 coelhos, separados em oito grupos, com oito animais. Os grupos foram: Gprpc (PRP, sem esponja cirúrgica, enxerto camada), Gprpce (PRP, esponja cirúrgica, enxerto camada), Gcc (solução fisiológica 0,9%, sem esponja cirúrgica, enxerto camada), Gcce (solução fisiológica 0,9%, esponjas cirúrgicas, enxerto camada), Gprpm (PRP, sem esponja cirúrgica, enxerto malha), Gprpme (PRP, esponja cirúrgica, enxerto malha), Gcm (solução fisiológica 0,9%, sem esponja cirúrgica, enxerto malha) e Gcce (solução fisiológica 0,9%, esponjas cirúrgicas, malha). Os dados foram analisados pelo teste t emparelhado, Kruskal-Wallis, análise de variância, e teste de Tukey (P<0,05). Edema e exsudato presente com três e sete dias (P=0,03 e P=0,0049); coloração cianótica no 14º dia (P=0,0001); aspecto cosmético bom no sétimo e no 14º dia (P=0,00026 e P=0,0001); presença de células mononucleares (P=0,01) e polimorfonucleares (P=0,01); proliferação de fibroblastos discreta (P=0,01); colagenização intensa (P=0,05); hemorragia discreta (P=0,007); ausência de diferença significativa em necrose e reepitelização (P=0,2928 e P=0,1). Conclui-se que o emprego do PRP gel em enxertos cutâneos associando esponjas cirúrgicas como curativo compressivo favorece sua integração ao leito receptor sem a presença prévia de tecido de granulação.(AU)

ABSTRACT

The aim of this work is study the laparoscopic ovum pick-up (LapOPU) technique in spotted paca, describing surgery details, complications and oocyte recovery rate. Nine healthy adult non-pregnant captive females were used, in a total of 39 procedures. When the surgical plane of anaesthesia was achieved, the females were positioned at 20º Trendelenburg. Three 6mm trocars were placed on right and left inguinal and hypogastric regions. Abdomen was inflated with CO2 and the intra-abdominal pressure was stablished in 10mmHg. Follicular punctures were performed moving the ovaries with atraumatic forceps. For punctures, an 18-gauge 3.5 inch long needle attached to a vacuum system with pressure not exceeding 65mmHg was used. Oocytes were recovered into 50mL centrifuge tubes with media composed of PBS supplemented with 10 IU/mL of heparin and kept at 36°C. R Software was used for statistical analysis. Data normality distribution (Shapiro test) and variances homoscedasticity (Bartlett test) were tested and descriptive statistics (mean±SD) was used to present the results. It was only possible to perform LapOPU in 30 of 39 laparoscopies (76.92%). The surgical total time was 37.34 ± 18.53 minutes. The total number of visualized follicles, aspirated follicles, and retrieved oocytes were 502, 415, and 155, respectively. And the same parameters per animal were: 14.34 ± 12.23, 11.86 ± 10.03, and 4.43 ± 4.69 respectively. Oocyte recovery rate was 32.56 ± 27.32%. In conclusion, caudal positioning of portals with slight triangulation allows good viewing of the abdominal cavity and eases the manipulation of the ovaries. Thus this described LapOPU technique is feasible in spotted paca and easy to perform.(AU)


Objetiva-se, com este trabalho, estudar a técnica de aspiração folicular por videolaparoscopia (LapOPU) em pacas, descrevendo detalhes do procedimento cirúrgico, complicações e taxa de recuperação oocitária. Para isso, foram utilizadas nove fêmeas, saudáveis, adultas, não gestantes e mantidas em cativeiro, totalizando 39 procedimentos. Quandoo plano anestésico cirúrgico foi alcançado, as fêmeas foram posicionadas em Trendelenburg com 20º de angulação. Três trocáteres foram colocados nas regiões inguinais direita e esquerda e hipogástrica. O Abdômen foi insuflado com CO2, e a pressão intra-abdominal foi mantida em 10mmHg. Punções foliculares foram realizadas manipulando-se os ovários com pinças atraumáticas. Para aspirações foliculares, usou-se agulha de 18G com bisel curto acoplado ao sistema de vácuo com pressão não excedendo 65mmHg. Oócitos foram recuperados em tubos de centrifugação de 50mL contendo meio composto de PBS suplementado com 10UI/mL de heparina e mantidos a 36ºC. Usou-se software R para análise estatística. Testaram-se a distribuição normal dos dados (teste de Shapiro) e a homocedasticidade das variâncias (teste de Barlett) e se usaram estatísticas descritivas (média±DP) para apresentar os resultados. Das 39 videolaparoscopias, só foi possível realizar LapOPUem 30 delas (76,92%). O tempo cirúrgico total das LapOPU foi de 37,34 ± 18,53 minutos. Os números totais de folículos visualizados, folículos aspirados e oócitos recuperados foram: 502, 415 e 155, respectivamente. E os mesmos parâmetros por animal foram: 14,34 ± 12,23, 11,86 ± 10,03 e 4,43 ± 4,69, respectivamente. A taxa de recuperação foi de 32,56 ± 27,32%. Assim, conclui-se que o posicionamento caudal de portais, com ligeira triangulação, permite uma boa visualização da cavidade abdominal e facilita a manipulação dos ovários, sendo essa técnica de LapOPU viável em pacas e de fácil execução.(AU)

RESUMO O presente trabalho descreve a composição físico-química, a avaliação da atividade antioxidante pelo método de sequestro de radicais livres DPPH, determinação de flavonoides e de fenóis totais dos extratos etanol, acetona e aquoso de Plectranthus barbatus Andr. (Lamiaceae), desidratadas em secador solar e em estufa de circulação de ar a 60 oC. Os valores de atividade de água encontrados para as duas secagens foram inferiores ao mínimo necessário para o crescimento e produção de toxina de patógenos de importância alimentar. Os resultados das análises físico-químicas demonstraram que ambos os processos mostraram-se eficientes na desidratação de P. barbatus. Os resultados demonstraram que os extratos acetona (estufa) e etanol (estufa e secador solar) foram os que apresentaram maior conteúdo de fenóis totais. O extrato etanólico (estufa) apresentou maior quantidade de flavonoides e melhor potencial antioxidante (IC50 = 75,71 ± 10,57 µg mL-1).


ABSTRACT This paper describes the physicochemical composition, the evaluation of the antioxidant activity by free DPPH radicals using the scavenging method, the determination of flavonoids and total phenolic compounds of ethanol, acetone and aqueous extracts of the medicinal plant Plectranthus barbatus Andr. (Lamiaceae), dehydrated in solar dryer and circulation oven at 60 °C. Water activity rates for two drying methods were below the minimum necessary for growth and toxin production of important food pathogens. Physicochemical results showed that both processes were effective in the dehydration of P. barbatus. The results demonstrated that the acetone (over) and ethanol (over and solar dryer) extracts showed the highest content of total phenols. The ethanol extract (over) showed the highest amount of flavonoids and better antioxidant activity (75.71 ± 10.57 µg L-1).

RESUMO As plantas produzem uma grande variedade de metabólitos secundários que frequentemente são relacionados a mecanismos de proteção da planta contra predadores e patógenos. As espécies tóxicas são aquelas capazes de produzirem compostos que podem causar alterações metabólicas prejudiciais ao homem e aos animais. A toxicidade apresentada por uma espécie vegetal pode estar relacionada a fatores associados ao indivíduo, à planta, ao modo de exposição e a questões ambientais. A intoxicação, aguda ou crônica, causada por plantas é difícil de ser diagnosticada assim como a associação entre os sintomas e o consumo e/ou contato com algumas espécies é difícil de ser estabelecida. No âmbito da saúde pública, as intoxicações causadas por plantas possuem impacto expressivo. No Brasil foram registrados 1026 casos em 2012, sendo que a maior parte deles ocorreu com crianças de 0 a 4 anos, de acordo com os dados do SINITOX. O objetivo deste trabalho foi realizar um levantamento bibliográfico de espécies vegetais, brasileiras e exóticas aclimatadas, citadas como tóxicas apesar de serem utilizadas com fins ornamentais e medicinais.


ABSTRACT Plants produce a wide variety of secondary metabolites, which are frequently related to a plant’s protective mechanism against predators and pathogens. Toxic species are those capable of producing compounds that can cause metabolic changes harmful to humans and animals. The toxicity of plant species can be associated with aspects related to the individual, the plant, the manner of exposure, and to environmental issues. Acute or chronic intoxication caused by plants is difficult to diagnose and the association between the symptoms and the consumption of and/or contact with plants is hard to establish. In the public health sector, intoxications caused by plants have a wide impact. In Brazil, 1,026 cases were registered in 2012, most of which occurred with children between the ages of 0 to 4 years, according to data reported by SINITOX. The purpose of this study was to perform a bibliographic survey of Brazilian or acclimatized exotic plant species, which have been reported as toxic even though they are used for ornamental or medicinal purposes.

ABSTRACT

O fruto de noni (Morinda citrifolia L.) é consumido há milênios na medicina popular polinésia devido aos benefícios nutricionais e terapêuticos. O consumo de noni em outros países, incluindo o Brasil, cresceu vertiginosamente nos últimos anos em decorrência das atividades biológicas atribuídas a ingestão do suco da fruta, principalmente pela propriedade anticâncer. Contudo, a composição química da planta, que está relacionada com suas propriedades biológicas, é determinada pelo seu local de origem, e por influência do clima e do solo onde é cultivada. Neste sentido, este trabalho teve como objetivo analisar a polpa extraída de frutos maduros de noni cultivados em Maringá-PR. A análise da polpa in natura apresentou 89,16% de umidade, 0,75% de cinzas, 2,10% de proteínas, 2,19% de lipídios e 5,81% de carboidratos. Dos compostos bioativos, foram analisados antocianinas (1,39 mg.100 g-1 polpa), flavonoides amarelos (13,01 mg.100 g-1 polpa), carotenóides (0,45 mg.100 g-1 polpa) e vitamina C (12,16 mg.100 g-1 polpa). Para fenólicos totais e atividade antioxidante foram preparados diferentes extratos, sendo que os maiores teores de fenólicos totais foram encontrados no extrato aquoso (1143,56 mg equivalente de ácido gálico (EAG).100 g-1), seguido do extrato etanólico (966,96 mg EAG.100 g-1), metanol/acetona (820,88 mg EAG.100 g-1) e metanólico (306,33 mg EAG.100 g-1). Os melhores resultados para antioxidantes, determinado pelo EC50 - concentração do extrato necessária para reduzir 50% do radical DPPH, foram encontrados nos extratos metanol/acetona (EC50 de 25,18 mg.mL-1) e metanólico (EC50 de 25,96 mg.mL-1). A atividade antioxidante dos frutos pode estar relacionada com o conteúdo de vitamina C, uma vez que os extratos com um menor conteúdo de fenóis totais foram aqueles que apresentaram menores valores de EC50.


The noni fruit (Morinda citrifolia L.) has been consumed for millennia in the Polynesian folk medicine because of its nutritional and therapeutic benefits. The consumption of the noni fruit in other countries, including Brazil, has increased in recent years because of the biological activities attributed to the ingestion of noni juice, especially the anticancer effect. However, the chemical composition of the plant, which is related to its biological properties, is determined by its geographical origin and is influenced by climate and soil. Therefore, this study aimed to analyze the pulp extracted from noni fruit grown in Maringá, state of Paraná, Brazil. The fresh pulp analysis showed 89.16% of moisture, 0.75% of ash, 2.10% of protein, 2.19% of fat and 5.81% of carbohydrates. The bioactive compounds analyzed were anthocyanins (1.39 mg.100 g-1 pulp), yellow flavonoids (13.01 mg.100 g-1 pulp), carotenoids (0.45 mg.100 g-1 pulp) and vitamin C (12.16 mg.100 g-1 pulp). For the analysis of total phenols and antioxidant activity, different extracts were prepared. The highest total phenolic contents were found in the aqueous extract (1143.56 mg EAG.100 g-1), followed by the ethanol extract (966.96 mg EAG.100 g-1), methanol/acetone extract (820.88 mg EAG.100 g-1) and methanol extract (306.33 mg EAG.100 g-1). The results for the antioxidant capacity were determined by the EC50, which is concentration of extract required to reduce 50% of the DPPH radical. The best results for the antioxidant capacity were found in the methanol/acetone extract (EC50 of 25.18 mg.mL-1) and in the methanol extract (EC50 of 25.96 mg.mL-1). The antioxidant activity of the fruit may be related to its vitamin C content, since the extracts with lower total phenolic content were those that had lower EC50 values.

ABSTRACT

This experiment was designed to evaluate the effects of the inclusion of detoxified castor bean meal (CM) content on the chemical-bromatological composition, as well as the fermentation characteristics and dry matter losses of sugarcane silage. The treatments consisted of four levels (0, 7, 14 and 21% natural matter) of addition of castor bean meal. The design was completely randomized, with five replicates. The material was ensiled in PVC silos of 50-cm in height and 10-cm in diameter, which were opened after 60 days. The density of fodder maintained in the silos was equal to 750kg of natural matter/m3. In order to quantify the gas and effluent losses, the silos were weighed at the beginning and at the end of the experiment. The addition of castor bean meal has increased the levels of DM and crude protein and reduced the levels of neutral detergent fiber, N-NH3, ethanol, and gas and effluent losses from silages, but did not affect pH values. During ensiling, alcoholic fermentation was controlled with the inclusion of the additive.


O experimento foi desenvolvido para avaliar os efeitos da inclusão de níveis de farelo de mamona destoxificado sobre a composição químico-bromatológica, as características fermentativas e as perdas de matéria seca da silagem de cana-de-açúcar. Os tratamentos consistiam em quatro níveis (0, 7, 14 e 21% da matéria natural) de inclusão do farelo de mamona. O delineamento foi inteiramente ao acaso, com cinco repetições. Foram usados silos de PVC, com 50cm de altura e 10cm de diâmetro, para a produção das silagens, que foram abertos após 60 dias. A densidade de forragem nos silos foi correspondente a 750kg de matéria natural/m3. Os silos foram pesados no início e ao final do período experimental para quantificar as perdas por gases e efluentes. A inclusão de farelo de mamona elevou os teores de MS e proteína bruta e reduziu os teores de fibra em detergente neutro, N-NH3, etanol e as perdas por gases e efluentes das silagens, não ocasionando efeito nos valores de pH. A fermentação alcoólica durante a ensilagem foi controlada com a inclusão do aditivo.

ABSTRACT

Objetivou-se descrever os achados clínicos e ultrassonográficos de uma cabra leiteira apresentando mastite gangrenosa. Com a ultrassonografia convencional, observaram-se alterações consistentes com o avanço do quadro clínico, como alterações na ecogenicidade do leite e parênquima mamário. A ultrassonografia Doppler revelou aumento da velocidade de pico sistólico nos três momentos estudados, passando de 15 para 17 e 29cm/s. Houve redução, também, dos valores do índice de resistência, de 0,71 para 0,61 e 0,43. O índice de pulsatividade sofreu redução entre o segundo e o terceiro dia de observação, passando de 0,98 para 0,71. Foi possível estabelecer relação direta entre os achados ultrassonográficos e a gravidade da doença. A ultrassonografia convencional e Doppler podem ser utilizadas para o estudo hemodinâmico do úbere de cabras leiteiras, favorecendo diagnóstico e prognóstico de alterações nesse órgão...

ABSTRACT

Dye exclusion tests are used to determine the number of live and dead cells. These assays are based on the principle that intact plasma membranes in live cells exclude specific dyes, whereas dead cells do not. Although widely used, the trypan blue (TB) exclusion assay has limitations. The dye can be incorporated by live cells after a short exposure time, and personal reliability, related to the expertise of the analyst, can affect the results. We propose an alternative assay for evaluating cell viability that combines the TB exclusion test and the high sensitivity of the flow cytometry technique. Previous studies have demonstrated the ability of TB to emit fluorescence when complexed with proteins. According to our results, TB/bovine serum albumin and TB/cytoplasmic protein complexes emit fluorescence at 660 nm, which is detectable by flow cytometry using a 650-nm low-pass band filter. TB at 0.002% (w/v) was defined as the optimum concentration for distinguishing unstained living cells from fluorescent dead cells, and fluorescence emission was stable for 30 min after cell treatment. Although previous studies have shown that TB promotes green fluorescence quenching, TB at 0.002% did not interfere with green fluorescence in human live T-cells stained with anti-CD3/fluorescein isothiocyanate (FITC) monoclonal antibody. We observed a high correlation between the percentage of propidium iodide+CD3/FITC+ and TB+CD3/FITC+ cells, as well as similar double-stained cell profiles in flow cytometry dot-plot graphs. Taken together, the results indicate that a TB exclusion assay by flow cytometry can be employed as an alternative tool for quick and reliable cell viability analysis.

ABSTRACT

Os objetivos desta pesquisa foram caracterizar as recusas por parte da população em permitir a borrifação de seus imóveis, em quatro regiões de Belo Horizonte, no período de 2006 a 2008, e relacionar estas recusas com o surgimento de casos de leishmaniose visceral, no mesmo local e período. Utilizaram-se as localizações onde se encontraram 27 casos humanos de leishmaniose visceral obtidos do SINAN, todos referentes a quatro áreas de abrangência da região noroeste de Belo Horizonte, no período de 2006 a 2008. De um total de 33.579 residências visitadas pelo controle químico vetorial, 9636 (28,70%) aceitaram a borrifação interna e a externa do domicílio, 20741 (61,77%) concordaram com a borrifação apenas do peridomicílio, 141 (0,42%) aceitaram apenas a borrifação do intradomicílio, 909 (2,70%) se encontraram fechadas no momento da borrifação e 2152 (6,41%) recusaram qualquer tipo de borrifação no imóvel. Na maioria das residências, o controle químico foi permitido apenas no peridomicílio. A diferença entre o perfil de recusas dos quarteirões contidos em uma área de 200 metros ao redor de um caso humano de leishmaniose visceral e o mesmo perfil dos quarteirões fora desta área foi significativa (P<0,05) em sete (25,92%) dos 27 casos da doença.


The purpose of this study was to characterize the population's refusal to allow their houses to be sprayed in four regions of Belo Horizonte, in the period from 2006 to 2008, and relate these denials with the emergence of cases of visceral leishmaniasis in the same location and time. We used the location of 27 human cases of visceral leishmaniasis obtained from SINAN, all data from four regions in the West Region of the city of Belo Horizonte, from 2006 to 2008. Of a total of 33,579 houses visited by the chemical vector control, 9636 (28.70%) accepted spraying inside and outside the home, 20741 (61.77%) agreed with spraying only the areas surrounding the property, 141 (0.42%) agreed only to spray indoors, 909 (2.70%) were found closed at the time of spraying, and 2152 (6.41%) refused any kind of spray at house. A chemical control vector was allowed only in surrounding areas of most of the houses studied. The difference between the profile of denials of the blocks contained in an area of 200 meters around a human case of VL, with the same profile of the blocks out of this area was significant (P<0,05) in 07 (25,92%) of 27 cases of the disease.

ABSTRACT

The effect of substitution of equine chorionic gonadotropin (eCG) by follicle stimulating hormone (FSH) in synchronization protocols of ovulation in dairy goats was evaluated. Twelve goats received intravaginal sponges impregnated with 60mg of medroxyprogesterone acetate (MAP) for 10 days. The sponges were removed and the animals were distributed into two groups (G): G1 (n=6) treated with 0.5mL of a synthetic analogue of PGF2 α and 100 IU of eCG for each 10kg weight, intramuscular injection (IM); and G2 (n=6) treated with 0.5mL of a synthetic analogue of PGF2α and 20mg of FSH (IM). All animals were monitored for estrus detection with aid of a ruffian after sponge removal. The ovarian dynamics were analyzed by ultrasound, since six hours after sponge removed. Each animal was analyzed in time elapsed of six hours until 12 hours after ovulation detection. For data analyses, the Wilcoxon test and variance analyses were used. There was not difference between the analyzed parameters (P>0.05). In this way, eCG can be replaced by FSH in synchronization protocols of ovulation in dairy goats.

Identificar fatores da linha de base preditores do alcance das metas do programa de intervenção no estilo de vida após 12 meses em população de nipo-brasileiros, empregando-se modelos de regressão logística ajustados. Em 2005, 321 participantes eram portadores de excesso de peso e houve maior chance [OR (IC95 por cento)] de alcance da meta de perda de peso após 12 meses entre mulheres [2,45 (1,33; 4,13)], indivíduos de maior idade [1,03 (1,00; 1,06)] e menor chance entre portadores de morbidades no início do estudo [0,33 (0,14; 0,77)]. Dos 261 indivíduos sedentários, o alcance da meta de atividades físicas foi inversamente relacionado ao exercício de atividades profissionais [0,40 (0,17; 0,95)]. Não se verificou fatores da linha de base associados ao alcance das metas do consumo de legumes, verduras e frutas e gorduras saturadas da dieta após 12 meses. Indivíduos de maior idade, mulheres, não portadores de morbidades e sem exercício de atividades profissionais na linha de base apresentaram maior chance de alcance das metas após 12 meses de intervenção no estilo de vida.


The aim of this study was to identify baseline factors associated with achieving goals after a 12-month lifestyle intervention program in a Japanese-Brazilian population, using adjusted logistic regression models. In 2005, 321 participants were overweight. The odds [OR (IC95 percent)] of reaching the goals after 12 months of intervention were directly related to female gender [2.35 (1.34, 4.13)] and older age [1.03 (1.00, 1.06)] and inversely related to baseline morbidity [0.33 (0.14, 0.77)]. Of the 261 sedentary individuals, achieving the goal for physical activity was inversely related to working [0.44 (0.17, 0.95)]. No baseline predictors were found for reaching the goal of fruit and vegetable consumption or saturated fat intake after 12 months. At baseline, women, older individuals, and individuals without diseases or not working showed increased odds of achieving the goals after 12 months of the lifestyle intervention.

ABSTRACT

The aim of the present study was to investigate the zooplankton community at six reservoirs in the state of Pernambuco (Brazil). Zooplankton assemblages were identified using current literature and quantitatively analysed under a microscope and stereomicroscope. Concurrently to the sampling of zooplanktonic and phytoplanktonic communities, in situ measurements of abiotic variables, such as water temperature, dissolved oxygen and pH, were determined using field probes and transparency was determined with a Secchi disk. Total phosphorus concentrations were used for the determination of the Trophic State Index. The reservoirs were classified between eutrophic and hypertrophic, oxygenated, with pH varying from slightly acid to alkaline, high temperatures and low water transparency. A total of 27 zooplankton taxa were identified. Phytoplankton was represented by Cyanophyta, Chlorophyta, Baccilariophyta and phytoflagellates. The highest richness of species was observed for Rotifera (17), followed by Crustacea (8), Protozoa (1) and Insecta (1). Rotifers shared quantitative importance with Crustacea, which were mainly represented by juvenile forms. Jazigo Reservoir presented the highest diversity and equitability. Lowest diversity and equitability were recorded at the Poço da Cruz and Mundaú reservoirs, respectively. Dissimilarity was detected between the environments studied regarding zooplankton composition and structure.


O objetivo deste trabalho foi estudar a estrutura e dinâmica do zooplâncton em seis reservatórios do Estado de Pernambuco. A comunidade zooplanctônica foi identificada com literatura atualizada e a densidade analisada sob microscópio e estereomicroscópio. Concomitantemente às coletas das comunidades zooplanctônicas e fitoplanctônicas, foram medidas in situ algumas variáveis abióticas como temperaturas da água e do ar, oxigênio dissolvido, condutividade elétrica, pH, turbidez, usando sondas de campo, transparência com disco de Secchi. As concentrações de fósforo total foram usadas para a determinação do Índice de Estado Trófico. Os reservatórios foram classificados entre eutróficos e hipertróficos, oxigenados, pH levemente ácido a alcalino, temperaturas altas e baixos valores de transparência da água. O zooplâncton esteve composto por 27 táxons. O fitoplâncton foi representado pelas Cyanophyta, Chlorophyta, Baccilariophyta e Fitoflagelados. A maior riqueza de espécies foi observada para Rotifera, (17), seguida por Crustacea (8), Protozoa (1) e Insecta (1). Rotifera compartilhou a importância quantitativa com os Crustacea, estes representados principalmente pelas formas jovens. No reservatório de Jazigo, foram obtidos os maiores valores para diversidade e equitabilidade. Os valores mais baixos foram registrados em Poço da Cruz e Mundaú, respectivamente. Foi detectada dissimilaridade entre os ambientes estudados quanto à composição e estrutura do zooplâncton.
